Along with the spermogram, the determination of antisperm antibodies from sperm and sperm culture, the determination of the degree of sperm DNA fragmentation is an important genetic analysis in infertile couple investigations.
The spermogram gives an overview of the sperm count, mobility and morphology, but not the quality of the genetic material. Even if the patient has normozoospermia (when count, mobility and morphology are normal), sperm DNA can be fragmented. In this case, the Halosperm test is an effective method of assessing the quality of sperm DNA and thus provides us with valuable information and helps in choosing the optimal assisted human reproductive method (IUI, IVF, ICSI, PICSI).
Numerous studies have shown that there is a close link between the degree of sperm DNA fragmentation and oocyte fertilization rate, embryo quality and implantation, and the rate of miscarriages. The superiority of this method is manifested precisely by the correct selection of the assisted reproduction technique without any failed attempts delaying the final goal.
Determination of sperm DNA fragmentation is performed using the Halosperm® kit based on the Sperm Chromatin Dispersion (SCD) method, the sperm with non-fragmented DNA presenting a halo that can be viewed microscopically after coloring.
The Halosperm test is recommended in the following cases:
- Male infertility due to unknown causes;
- Repeated pregnancy losses;
- The man is over 40 years of age;
- Unhealthy lifestyle;
- Repeated exposure to chemical or toxic substances, radiation, high temperatures;
- Infectious diseases, varicose veins, testicular cancer, etc.
Results interpretation and related recommendations:
Fragmentation degree Recommendations:
<15% MILD DEGREE OF FRAGMENTATION
The percentage of sperm with DNA integrity is high, which allows natural fertilization (in the absence of other problems associated with infertility).
>15% – <25% MODERATE DEGREE OF FRAGMENTATION
The degree of fragmentation is moderate. The indication in this case is artificial insemination (in the absence of other problems associated with infertility).
>25 – <50% HIGH DEGREE OF FRAGMENTATION
The degree of fragmentation is significant, the recommendation being given for IVF, ICSI or even PICSI.
> 50% SEVERE DEGREE OF FRAGMENTATION
The degree of fragmentation is very high, with most sperm presenting an altered genetic material. In this case, the recommendation is given for IVF with PICSI.
The Halosperm test is mandatory prescribed in conjunction with the Spermogram.
Along with these two tests, international protocols also recommend:
Anti-spermatozoa (IgA / M / G, from seminal fluid);
Zinc (Zn) in seminal fluid.
